Follistatin is a peptide that originates from the body, but has been adapted into a new peptide that is helping build muscle and strength. Researchers hope this find will help treat muscle wastage diseases in the future. For researchers in this field searching for follistatin for sale check out our shop today!
What is Follistatin-344?
Follistatin-344 is an FDA approved small molecule drug. It is an inhibitor of the enzyme activin type 2 receptor. This enzyme is a member of the TGF beta superfamily of receptors and it is a regulator of BMP-9. Follistatin-344 blocks the biological action of activin A (and BMP9) which acts on target cells, including stem cells, with effects including cell proliferation, growth and differentiation. Clinical trials are being carried out to investigate the effect of follistatin-344 on bone regeneration, but there are no indications for use in the area of dentistry at present.
Follistatin 344 Cycle ?
Follistatin-344 is a new drug that is not being currently used in the dental field, but it is being used as a drug for the treatment of muscle wasting and to improve athletic performance.

In humans, follistatin-344 stimulates the differentiation of adipocytes. Follistatin-344 does not affect cell viability or adipocyte apoptosis. It stimulates the proliferation of mesenchymal stem cells and may act on the epithelium of gingival tissues. It is also being studied as a possible drug for treating muscle wasting.
Follistatin 344 Benefits
This approach allowed one to create the first mouse model of ovarian cancer using a novel conditional gain-of-function strategy. In this model, the first tumour developed in the ovaries. As reported by our group, the tumours grew rapidly. All mice died of tumour spread or cachexia. Importantly, it was found that, once formed, the tumour cells remained dependent on the presence of follistatin 344 for their growth. Thus, these tumours could be eliminated by administration of a therapeutic antisense against follistatin 344.
How to use Follistatin 344 In order to confirm this strategy as a valid therapy, a phase I study was initiated in which the therapeutic antisense oligonucleotides were used to block *Fst* transcription in ovarian cancer. The oligonucleotide sequence 5'-AGCAGCATGGCATGCATC-3' was designed and incorporated into a phosphorothioate-modified 2'-O-Me ribose backbone.
Follistatin Dosage
This sequence forms an intramolecular RNA duplex and also contains 2'-O-Me nucleotides in the middle of the duplex, which are expected to facilitate the dissociation of the duplex upon target RNA recognition. These design features make the oligonucleotide suitable for *in vitro* and *in vivo* use. As expected, follistatin 344 levels were reduced in the tumours and ascites in mice treated with the therapeutic *Fst* oligonucleotides. Moreover, the tumours were eliminated, and mice did not develop tumours or show any of the symptoms associated with tumour formation.
Follistatin 344 Protocol
In recent years, several publications have described the downregulation of FST in breast cancer \[[@B43-biomolecules-04-00458]\], head and neck squamous cell carcinoma \[[@B44-biomolecules-04-00458]\], glioblastoma \[[@B45-biomolecules-04-00458]\], and pancreatic \[[@B46-biomolecules-04-00458]\] and lung \[[@B47-biomolecules-04-00458]\] cancer.

However, *FST* gene knockdown did not result in increased tumour growth or a worse prognosis. The same was true for ovarian cancer \[[@B45-biomolecules-04-00458]\], as previously discussed. Thus, based on the findings of our studies and those of other investigators, it seems that FST is involved in the regulation of other types of cancers. The mechanism of FST downregulation remains to be elucidated. However, one possibility is the post-transcriptional destabilisation of the *FST* transcript through, for example, miRNA-induced mRNA degradation.
